Our institution is conducting a clinical trial of ASA404, an investigational drug to be used in combination with docetaxel to treat patients with locally advanced or metastatic Non–Small Cell Lung Cancer (NSCLC) who have progressed while on or following first-line cytotoxic chemotherapy. We are seeking referrals of appropriate patients for enrollment into this lung cancer study.
ASA404 is a novel Tumor-Vascular Disrupting Agent (Tumor-VDA) that inhibits tumor blood flow and causes tumor necrosis. Docetaxel alone has been demonstrated to increase overall survival as second-line treatment for patients with NSCLC and is approved worldwide for this indication. ASA404 has demonstrated preclinical synergy with taxanes, and data from phase II trials indicate that the addition of ASA404 may increase the efficacy of taxane-based therapy for lung cancer. This and the different toxicity profiles of ASA404 and docetaxel provide a compelling rationale for combination treatment.
The clinical study, called ATTRACT-2 (Antivascular Targeted Therapy: Researching ASA404 in Cancer Treatment), is a phase III clinical trial being conducted worldwide in order to demonstrate an overall survival advantage of ASA404 in combination with docetaxel for second-line treatment of advanced NSCLC. With an accrual target of 900 patients, this important study may help redefine the way NSCLC is treated.
